What is Spravato?

Spravato is a prescription nasal spray containing esketamine, a derivative of ketamine. It is FDA-approved for two specific indications:
  • Treatment-Resistant Depression (TRD): For adults who have not responded to at least two antidepressants taken at adequate dose and duration. Spravato is used in conjunction with an oral antidepressant.
  • Major Depressive Disorder with Acute Suicidal Thoughts or Actions: For adults in crisis who require immediate symptom relief. Spravato® is used alongside an oral antidepressant to stabilize mood rapidly.

Who is Spravato For?

Spravato® may be appropriate for adults who meet the following criteria:
  • Have a diagnosis of TRD and have not improved with at least two antidepressants.
Limitations of Use:
  • Spravato® is not indicated for use in children.
  • Hospitalization may still be necessary even if rapid symptom relief is observed.

How Does Spravato Work Differently?

Unlike traditional antidepressants that affect serotonin, dopamine, or norepinephrine, Spravato® works on the NMDA receptor—a different pathway involved in glutamate signaling. This is believed to restore synaptic connections in the brain, offering faster symptom relief for individuals with difficult-to-treat depression.
